Here we determine a high-quality genome sequence of Ramazzottius varieornatus, one of the most stress-tolerant tardigrade species. Excessive NUMTs or NUPTs were not detected. Genome and RNA-expression comparisons between H. dujardini, which is able to adopt its tun form only after exposure to gradual drying conditions, and R. varieornatus, which dessicates rapidly, revealed candidate genes and pathways that may play a role in tardigrades’ survival abilities.
